I think a certain few do.  Funaho seems to know something in episode 13.

Had Tsunami wanted the throne, she could have taken it at any time.  Tsunami is looking
for Tenchi and in the process, promised to protect the first ruler of Jurai's decendants.
During that time, there were several rulers on Jurai and Tsunami never
attempted to join with anyone.  Of course, she was destined to join with Sasami so
the fact that she did nothing prior means little.  Still, Tsunami isn't interested
in ruling Jurai.

Yes.  Aeka immediately recognized Tsunami's humanoid form in episode 9.  I believe
Tsunami appeared to Azusa in the novel "Jurai" in humanoid form when she rescued him
(have to check that though).  It appeared that Nobuyuki and Yosho recognized the
ghostly figure as Tsunami as well in episode 9.
